Coming from a Linux background, I was able to block certain hosts from accessing NFS shares. Is this possible with Windows 11 Home folder sharing? All of the guest VMs are Linux-based (using Hyper-V).

Can you expand on what your trying to do. I am reading this as you want to block specific hosts (VMs) from specific shares (or all shares on a given host)?

Exactly. Since I'm very familiar with NFS, I would prefer using an NFS server solution. Do you know if this is possible via WSL?

Rights to shares can easily be administered thru the GUI.
If you want to block a system, use the Hosts file.
I am not sure for the Home version though.

I think I've been overthinking this, making the situation unnecessarily complicated. What I have currently setup is (4) shared folders in my home directory, 2 of which I would like to block access to all systems except one. Not even sure how I would do this with NFS, to be honest.

I will post a follow-up after I've decided exactly what I'm trying to accomplish here.

If we're talking about SMB shares then the answer is no you can not block a given host per share. SMB authn/z is user/group based.

If you'd rather do NFS I believe it is possible via WSL 2.0
Systemd is available Systemd support is now available in WSL!

You will also need to modify the network mode of WSL, normally it runs in NAT but can be setup for local LAN access
