If you mean: magnifier, you don't need to use any registry mod for that.
Settings > Ease of Access > Magnifier and then switch off will do.

Or pressing Windows key + Esc, does the same.

If you mean something else, please describe more specified.

Make a backup of Magnify.exe and rename it to something like Magnify.exe.bak

You could try right clicking on C:\Windows\System32\Magnify.exe and removing ALL permissions from everybody for the file.
See if Windows can still open it?

If it works, there’s a good chance it will revert back after an update or running SFC or something.

    …still on a horse.
You could install AutoHotKey 2.0 (freeware) and make this script:

; Test for switching off magnifier shortcuts
<#+::
<#=::
<#NumpadAdd::
>#+::
>#=::
>#NumpadAdd:: return

After that, the left and right Windows key, in combination with either = or + (shift =) or numpad + key will not enable the magnifier anymore.

Just did the test: does function.

AutoHotKey is a very useful program anyway. Just a bit complicated to use, but they have an excellent help file.
