I take the view to do them separately.
Firstly, that way if one fails, you do not lose the whole backup.
Also, you may not need to backup each drive to the same frequency.
TBH I only backup my OS drive with Reflect. There are better ways really to backup data drives which will backup in real time, only making backups to changed data which much reduces backup time.

As cerberus says, do them individually as this gives you more flexibility.
I have C full weekly with differential daily
D for documents weekly with differential daily
With the above i can restore C and not have to worry about losing emails etc
