Yeah, it should be blocking the actual executable, regardless of protocol or port, at this point. So it bears investigating why your setup is not outright blocking it. I suspect it might have to do with the default monitoring for your system being set to "always ask".
Typically, the strongest security is to deny all , and then selectively allow the ones that you want to get through, and since I haven't actually used GlassWire itself, I suspect that might be a workable solution - if you don't mind allowing all the stuff that normally goes through perfectly fine being (temporarily) blocked until you make an exception to allow it through.
I'd consult their documentation and help files (or forum, if they have one) and see if someone else has already run into this issue specifically with SharePoint. You might find a good happy medium that allows you to block it completely and otherwise maintain your current methodology for connectivity and setup using GlassWire.
