No, it's preferable if it's on. I've seen anecdotal reports saying it's actually running even though the switch is off and it says it isn't, but I have trouble believing that. Anyway, go ahead and enable it.

I just checked two of my Windows 11 computers, and Core Isolation is turned off.
I don't remember turning it off.

Also. On one computer, Microsoft Vulnerable Driver Blocklist was also turned off.
Both computers had Local Security Authority Protection turned on.
